Service

In vitro - Bioreactor
Bioreactor cartridges provide the best alternative to using animals for custom monoclonal and hybridoma antibody production. One bioreactor cartridge can produce as much antibody as 10 to 12 ascites mice. In addition, the custom monoclonal and hybridoma antibodies in production in bioreactors contain fewer mouse proteins and nonspecific antibodies. Another advantage of our custom hybridoma and monoclonal antibody production is that some hybridoma cells do not grow and produce well in mice. In these cases the bioreactor system is an excellent animal-free alternative for custom bulk antibody production. 100 mL is the standard service.


Step 1: Materials needed from client for custom monoclonal and hybridoma antibody production: 1 x 106 - 1 x 107 hybridoma cells, viable, shipped on dry ice. IPA cultures these prior to inoculation of flask.

Step 2: The membrane compartment is inoculated with 50 to 500 million cells, depending on the size of bioreactor, and incubated for about a week.

Step 3: Supernatant rich with antibodies and cells (7.5 mL) are harvested twice per week (Mon and Thurs) after rapid growth is established. for a total of 15 mL per week.

7 Weeks

Step 4: The cells are removed by centrifugation and filtration and the antibody-containing supernatant is ready for use or further purification or modification.

 

Yield: 100 mL (25-50 mg, 100 in a perfect world), 1 mg/mL upper limit.

In vivo - Ascites fluid
Ascites fluid is often an ideal medium for the reliable production of custom monoclonal, hybridoma antibodies. Clients can take advantage of IPA?s full range of small and large scale custom ascites hybridoma and monoclonal antibody production services in diverse mouse and rat strains. Live or frozen cells may be sent to IPA for custom ascites, monoclonal and hybridoma antibody production. Traditionally, Antibody containing ascites fluid is then tapped from each mouse up to three times.


Step 1: Receipt and Expansion:
Materials needed from client for custom monoclonal, hybridoma antibody production: 5 x 106 hybridoma cells, viable, shipped on dry ice. Upon receipt, all cell lines are assigned an Accession Number for tracking purposes. The cell line is transferred to a quarantine area where it is grown in culture and expanded to create cells ready for inoculation.

Step 2: Inoculation
Monoclonal antibodies are produced in bulk by implanting hybridoma cells into the peritoneal cavity of a primed mouse. Custom Ascites, Hybridoma and Monoclonal antibody production is initiated under cGMP guidelines.

Step 3: Production Harvest
Ascites is collected on wet ice and centrifuged to remove cellular and lipid material. The clarified ascites is placed into sterile containers, labeled, and immediately frozen.

Yield: 5 mice provide 15 - 50 mL total (10 mg/mL average mouse ascites concentration).

Sample Submission

In Vitro (Integra BioReactor):

1 x 106 - 1 x 107 hybridoma cells, viable, shipped on dry ice. IPA cultures these prior to inoculation of flask.

In Vivo (ascites):

5 x 106 hybridoma cells, viable, shipped on dry ice. The cell line is grown in culture and expanded to create cells ready for inoculation.


Confidentiality

IPA maintains all custom hybridoma and monoclonal antibody production information under the strictest confidentiality. All information and/or materials supplied by the client will be used as directed. Upon completion of the custom hybridoma or monoclonal antibody production project, all materials will be either returned or disposed of as directed by the client. IPA is willing to execute confidentiality agreements with its clients.
