ImmunoPrecise has proven expertise in developing custom reagents for use in Mass Spectrometry applications and has released two new antibody products for use in absolute quantitation. The large-scale systematic quantitation of proteins is an important component of proteomics and has contributed to the emergence of the new fields of systems biology and molecular-based medicine. Quantitative proteomics will provide new insights into biological function, facilitate the identification of diagnostic and surrogate disease markers, and contribute to the discovery of proteins as therapeutic targets.  Mass spectrometry (MS) coupled with shotgun style experiments has proven to be a powerful tool in the study of proteomes, but currently available technologies provide only relative quantitation information (rather than absolute). It will take more targeted approaches and specialized purpose-built reagents to truly expose the rich complexity of protein expression, modification, and interaction in biological systems. Immunoreagents for MS developed by ImmunoPrecise will facilitate larger, more sensitive, multiplexed studies that will advance protein diagnostics to the point of truly quantitative and absolutely specific assays.

 

Service. mAbs for mass spectrometry includes:

 

It starts with analysis of your target protein to identify candidate sequences and synthesize two different proteotypic peptides (based on trypsin digestion) compatible with quantitative mass spectrometry based research (free of residues subject to modification etc).

 

Once the two targets (to allow validation) are developed we use our Rapid Prime mAb development strategy and specialized high-throughput high-affinity screening methods to identify the best mAb candidates for MS use.

 

To assess the antibodies’ performance as quantitative pull down reagents and their compatibility and utility in mass spec analyses we test the mAb's ability to specifically recognize its target peptide and retain it during the process of mass spectrometric analysis

Thyroglobulin. A pair of monoclonal antibody reagents that each specifically capture a distinct trypsin proteotypic peptide. The antibodies were developed to capture the same tryptic peptides that would be generated in a typical sample preparation for MALDI-TOF or LC/MS/MS experiments.
